玉米中一种新的蛋白激酶电子克隆与序列分析

【摘要】 目的:电子克隆玉米中一种新的蛋白激酶基因。方法:以拟南芥中一个蛋白激酶的氨基酸序列为探针,对玉米的EST数据库进行同源性检索和筛选并克隆。结果:序列分析显示该cDNA长1121bp,有一个450bp的开放阅读框,编码149个氨基酸,且具有保守苏氨酸/丝氨酸蛋白激酶结构域和TEY基元,说明所克隆的cDNA序列为玉米的MAPK全长cDNA。结论:所克隆的cDNA序列为玉米的MAPK全长cDNA。

【Abstract】 Objective:A novel corn(Zea mays) protein kinase gene was cloned In silico cloning.Methods: A novel corn(Zea mays) protein kinase gene was cloned by blasting the corn EST database with the Arabidopsis thaliana kinase protein as a querying probe.Results: Sequence analysis showed that the in silico cloned cDNA is 1 121 base pairs long with an open reading frame(ORF) containing 450 nucleotides encoding a protein of 149 amino acids.And it have the conserved Serine/Threonine protein kinases catalytic domain and MAPK motif TEY.All these results indicated that the in silico cloned cDNA is corn MAPK gene.Conclusion: The in silico cloned cDNA is corn MAPK gene.
