白芨胶载外源性rhEGF对创面中羟脯氨酸含量的影响

【摘要】 目的观察中药白芨胶(BSG)作为外源性重组人表皮生长因子(rhEGF)的载体,在兔背部的创面愈合过程中对羟脯氨酸(HYP)含量的影响,并探讨其机制。方法选取健康家兔40只,体重2.5~3.0kg,雌雄各半。随机分为盐水对照(SW)、BSG、rhEGF、BSG+rhEGF四组,于脊柱的两侧其正中旁开2.0cm处手术制作皮肤全层缺损创面各3个,面积约2.0cm×2.0cm。分别用浸透上述四种液体的纱布外敷创面,隔日换药1次。于外敷后第3、5、7、10天取材,行HYP含量的测定。结果四组的HYP含量于外敷后第3天均较低,以后逐渐递增,至第10天达较高水平。在同一时间段上,BSG+rhEGF组的HYP含量显著高于其余三组(P<0.05),BSG组略高于rhEGF组(P>0.05),但三组均显著高于SW组(P<0.05)。结论BSG+rhEGF能促进创面中HYP合成的增加,从而缩短了创面的愈合时间,提高了愈合质量。

【Abstract】 Objective To observe the effect of bletilia striata gelatin as a vector of recombinant human epidermal growth factor (rhEGF) on hydroxyproline contents in wounds. Methods Forty healthy rabbits weighting apporoximately 2.5~3.0 kg (male and female in half) were randomly divided into 4 groups which were administrated with SW, BSG, rhEGF and BSG+rhEGF respectively. After the backs of these rabbits were sterilized by general iodophors, the 3 full thickness skin defects (2.0 cm×2.0 cm) were created on the bilateral spinal column, which were 2.0 cm to the defects. The defects were covered with the gauzes which were soaked with the four kinds of physic liquor mentioned-above. Chang dressings were performed every other day. The tissues which were 2.0 cm outside the defects edge were taken on day 3, 5, 7, 10 respectively and the contents of hydroxyproline were determined. Results The cotents of hydroxyproline in defective tissues were lower on day 3, and were higher at day 10 by increasing gradually. At the same detectational period, the contents the hydroxyproline in BSG+rhEGF group were significantly higher than the other groups (P<0.05), the BSG group was gently higher than rhEGF group (P>0.05) but three experimental groups were significantly higher than SW group (P<0.05). Conclusion BSG+rhEGF could promote the synthesis of hydroxyproline in defects to shorten healing time and raise healing quality.
