VEGF在慢性硬膜下血肿液及其外膜中的表达

【摘要】 目的探讨血肿液及其外膜血管内皮生长因子(VEGF)的表达与慢性硬膜下血肿(CSDH)发生发展的关系。方法利用酶联免疫吸附分析法(ELISA)对CSDH患者血肿液和血清中VEGF的含量进行测定,利用免疫组织化学方法观察CSDH血肿外膜的VEGF蛋白表达,并根据F-Ⅷ因子在血管内皮中的表达情况进行微血管密度(MVD)的计数,利用电镜观察血肿外膜的超微结构,分析在CSDH中VEGF对血肿外膜毛细血管的生成作用及血肿外膜中毛细血管的形态学改变。结果血肿液中VEGF的含量明显高于血清中的含量(P<0.01),血肿外膜中可见明显增生的毛细血管和大量的炎症细胞,VEGF与F-Ⅷ因子均呈强阳性染色。电镜结果显示血肿外膜中存在大量新生毛细血管,血管通透性增高。结论VEGF与CSDH血肿外膜的毛细血管生成及血管通透性增高密切相关。

【Abstract】 Objective To probe vascular endothelial growth factor’s(VEGF)influence on the expression and the advance of chronic subdural hematoma(CSDH).Methods The concentrations of VEGF in hematoma fluid and serum were determined by enzyme-linked immunosorbent assay(ELASA)in patients with CSDH.VEGF protein expression and microvessel density(MVD)counting on outer membrane and duramater of CSDH in the different cases were observed by immunohistochemical method.VEGF’s capillary producing function on outer membrane and capillary’s morphological changes in outer membrane in CSDH were analyzed by means of observing outer membrane with electromicroscope.Results The concentrations of VEGF were significantly increased in the hematoma fluid as compared with that in the serum(P<0.01).VEGF displaying clear positive staining in outer membrane and positive staining in outer membrane were very obvious different to that in duramater(P<0.001),which presented obvious difference than that in the control group(P<0.001).MVD counting in outer membrane was obvious different to that of duramater and the control group(P<0.001).VEGF was obviously related to MVD counting(P<0.001).There were plenty of newly reproductive capillary in outer membrane,which induced the improvement of vascular ventilating function.Conclusion VEGF is close related to capillary producing on the outer membrane.
