血红素加氧酶1在新疆卡波氏肉瘤组织中的表达及意义
Expression of Heme Oxygenase-1 in Kaposi’s Sarcoma
【摘要】 目的探讨血红素加氧酶1(heme oxygenase-1,HO-1)和卡波氏肉瘤(Kaposi’s Sarcoma,KS)病因的相关性。方法用Real Time Quantitative PCR(简称RQ-PCR)方法检测7例KS组织中HO-1表达,同时分别以相应瘤旁正常组织和7例正常人正常组织作为对照组。首先抽提7例KS组织、7例瘤旁正常组织和7例正常人正常组织总RNA。以总RNA为模板,逆转录成总cDNA。以总cDNA或目的基因质粒为模板,在Smart Cycler全自动荧光定量PCR分析仪上同时扩增HO-1和GAPDH基因的标准曲线和检测样本中HO-1和GAPDH的表达。结果在KS组织中HO-1表达水平明显高于瘤旁正常组织(t=4.074,P<0.01);也明显高于对照组(t=2.731,P<0.05),并且结果有统计学意义。结论卡波氏肉瘤HO-1表达明显高于瘤旁正常组织和对照组,并且结果有统计学意义,说明HO-1在KS的发病中起到一定作用,具有重要相关性。
【Abstract】 To study the relationship between HO-1 and etiopathogenisis of Kaposi’s sarcoma(KS).Quantitative RT-PCR was performed to detect for expression of HO-1.First,RNA was isolated using TRIZOL in the KS group,the normal skin group of KS and the control group.And then,quantification of RNA was performed by a 2-step method.cDNA was synthesized in the first place.HO-1 and GAPDH were synthesized by cDNA.In the second place,quantitative RT-PCR was performed on a Smart Cycler Sequence Detection System under standard reaction conditions.From the results:expression levels of HO-1 in KS tissues was apparently higher than the normal skin group of KS(t=4.074,p<0.01),and also apparently higher than those of control group(t=2.731,p<0.05).There was a significant meaning in the results.So we come to the conclusion that HO-1 was overexpression in KS with significance,compared with the two controls.The results suggest that HO-1 was associated with development of KS.
