胶体金标记苯丙胺单克隆抗体免疫试剂盒研究
Study on the Immunochromatographic Test for the Detection of Amphetamine
【摘要】 目的建立一种准确、快速、简便的检测尿液中苯丙胺(AMP)的胶体金免疫层析技术。方法采用柠檬酸三钠还原法制备胶体金颗粒,标记抗AMP单抗,将AMP—BSA抗原固相于硝酸纤维素膜上,制备胶体金免疫层析测试条。通过尿液、血液和唾液中的可能存在的苯丙胺成分与测试条上的苯丙胺-BSA完全抗原竞争结合有限的单抗结合位点,来判定检测结果。结果用ICT法和GC/MS检测217份尿样,本法检测阈值为1000ng/mL,特异性为99.17%,准确性为99.54%。结论ICT法检测尿液中的AMP特异性强,灵敏度高、简便快速、无需特殊仪器设备,具有广泛应用价值。
【Abstract】 Objective To develop an accurate,rapid and simple colloidal gold immuno-chromatographic test(ICT) for the detection of amphetamine in urine.Methods Colloidal gold was prepared by reduction of HAuCl4 with trisodium citric acid and conjugated with anti-amphetamine monoclonal antibody.Amphetamine-BSA conjugate was made and coated on nitrocellulose membrane to get the test.Outcome was detectable as a result of the competition for limited antibody binding sites between amphetamine-BSA and free amphetamine which may be present in urine.Results A total of 217 urine samples were tested for amphetamine by ICT and GC/MS.The cut-off value was 1000ng/mL,specificity was 99.17% respectively and the accuracy was 99.54%.Conclusion ICT is a sensitive,rapid and simple assay for detecting amphetamine in urine.
