S100A9、HSP70在同步放化疗敏感性不同宫颈癌中表达的实验研究

【摘要】 目的:观察S100A9、HSP70在宫颈癌组织中的表达及其与同步放化疗敏感性的关系。方法:收集进行治疗前中晚期宫颈癌组织标本。放化疗后,根据WHO实体瘤疗效判断标准,从收集的标本中选10例,高敏感组(5例)和低敏感组(5例)。提取组织总蛋白,进行双向凝胶电泳和质谱分析鉴定差异表达蛋白,并用Western Blot进行验证;免疫组织化学方法检测部分差异表达蛋白的表达意义,其中高敏感组60例,低敏感组35例。结果:建立了宫颈癌同步放化疗高敏感组和低敏感组的双向凝胶电泳图谱,鉴定得到高敏感组中高表达的S100A9蛋白,以及低表达的HSP70蛋白。Western Blot及免疫组化结果显示S100A9在高敏感组中的表达强度和表达率均高于低敏感组,HSP70在低敏感组中的表达强度及表达率高于高敏感组。结论:S100A9和HSP70的表达与宫颈癌同步放化疗敏感性有关。

【Abstract】 Objective:To observe S100A9 and HSP70 expression in cervical carcinoma tissues with different concurrent radiochemotherapy sensitivity and its significance.Methods:The fresh carcinoma tissues were collected from 10 untreated cervical carcinoma patients.The tissues were classified into two groups: high sensitivity group(HS) and low sensitivity group(LS),according to their response to concurrent radiochemotherapy.Firstly,protein separating was performed by using two-dimensional gel electrophoresis(2-DE) The peptide mass fingerprintings(PMF) was acquired by matrix assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS) and the proteins were identified by data searching.Differentially expression proteins were assayed by Western Blot and with immunohistochemical method in clinical samples including 60 high-sensitivity cases and 35 low-sensitivity cases.Results:2-DE gels were obtained.It showed that S100A9 expression was higher in HS group than in LS group,and HSP70 expression was higher in LS group than in HS group.The higher expression of S100A9 in HS group and HSP70 in LS groups were further confirmed by western blot and immunohistochemical method.Conclusions:S100A9 and HSP70 expression were associated with concurrent radiochemotherapy sensitivity in cervical carcinoma.
