GDF-9和BMP-15在PCOS患者卵母细胞和卵冠丘细胞的表达研究

【摘要】 目的:探讨卵源性因子生长分化因子-9(GDF-9)和骨形态发生蛋白-15(BMP-15)在行体外受精-胚胎移植PCOS患者卵母细胞和卵冠丘颗粒细胞(cumulus granulosa cell,CGC)中的表达情况。方法:按1:4比例收集因男方因素行ICSI后废弃的PCOS患者和对照组患者未成熟生发泡(germinal vesicle,GV)期卵母细胞或第一次减数分裂中期I(metaphase I,MI)期卵母细胞。运用免疫荧光结合激光扫描共聚焦显微镜(LCSM)对GV期卵母细胞GDF-9蛋白的表达进行检测,对MI期卵母细胞进行BMP-15蛋白表达的检测;应用免疫细胞化学方法检测两种因子在MII期CGC的表达。结果:GDF-9和BMP-15在PCOS患者和对照组患者卵母细胞的表达差异无显著。GDF-9在PCOS患者CGC的表达强度为5.01±0.83,显著低于对照组患者的8.88±1.52,P<0.05。BMP-15在PCOS患者CGC的表达与照组患者比较差异无显著。结论:PCOS患者促排卵治疗后卵母细胞GDF-9和BMP-15表达无差异,而CGC GDF-9表达低提示促排卵治疗改善了PCOS卵泡的生长,但可能导致过早黄素化。

【Abstract】 Objective:To evaluate the expression of growth differentiation factor-9(GDF-9) and bone morphogenetic protein 15(BMP-15) in oocytes and cumulus granulosa cells(CGC) of polycystic ovarian syndrome(PCOS) patients.Methods:Germinal vesicle(GV) or metaphase I(MI) stage oocytes of PCOS patients and controls which not suitable for ICSI were collected from our IVF center.Immunofluorescence combined with laser scanning confocal microscope(LCSM) was used to examine the expression of GDF-9 protein in 12 GV stage oocytes and expression of BMP-15 protein in MI stage oocytes.The expression of these two factors on MII stage CGC was analyzed with immunocytochemistry,simultaneously.Results:No statistic difference was found in the expression of GDF-9 and BMP-15 in the oocytes between PCOS patients and controls.However,the expression of GDF-9 in CGC of PCOS patients decreased significantly(8.88±1.52 vs 5.01±0.83,P<0.05).Conclusions:The normal expression of GDF-9 and BMP-15 protein in oocyte of PCOS patients implies that gonadotropin stimulating ovulation improves the follicle growth.The lower expression of GDF-9 in CGC of this group may be associated with the premature differentiation of CGC and folliculargenesis.
