自身免疫性甲状腺疾病Th1/Th2相关细胞因子不同极化方向漂移的研究

【摘要】 目的探讨自身免疫性甲状腺疾病(AITD)发病的始动原因,明确AITD Th1/Th2相关细胞因子群谱的特征。方法用逆转录酶链聚合反应方法检测IFN-γ、TNF-α、IL-2、IL-4、IL-6、IL-10等6种细胞因子的mRNA表达。结果与正常对照组相比,Graves病(GD)和桥本甲状腺炎(HT)细胞因子表达率明显增加;GD Th2类细胞因子IL-4和IL-6总表达频率明显高于Th1类细胞因子IFN-γ、TNF-α和IL-2表达频率,他巴唑治疗前后GD细胞因子Th2强势的表达特征没有明显变化,但总的细胞因子的表达频率下降;HT Th1类细胞因子IFN-γ、TNF-α和IL-2表达频率高于Th2类细胞因子IL-2、IL-4、IL-6、IL-10的表达频率。结论GD细胞因子表达向Th2方向漂移,以体液免疫为主;他巴唑治疗后的GD细胞因子谱没有明显改变,而HT细胞因子表达则呈现Th1强势,以细胞免疫为主。

【Abstract】 [Objective] Studying the initial pathogenesis of autoimmune thyroid disease(AITD),and clarifying the Th1/Th2 cytokine profile in AITD.[Methods] The gene expression of IFN-γ、TNF-α、IL-2、IL-4、IL-6 and IL-10 were detected respectively by the method of reverse transcription polymerase chain reaction(RT-PCR).[Results] Compared with the control,the expression rates of all cytokines were significantly elevated in Graves’ disease(GD) and Hashimoto’s thyroiditism(HT).In GD,the expression rates of Th2-like cytokines(IL-4、IL-6) were much higher than those of Th1-like cytokines(IFN-γ、TNF-α、IL-2),but no change happened after being treated with methimazole.In HT,the expression rates of cytokines(IFN-γ、TNF-α、IL-2) were higher than those of Th2-like cytokines(IL-4、IL-6、IL-10).[Conclusion] The predominant expression of Th2-like cytokines mainly mediating humoral immunity was found in GD,and the expression of cytokine profile was unchanged after being treated with methimazole,while a predominant expression of Th1-like cytokines mainly mediating cell immune responses was found in HT.
