长期口服家蚕生产的CTB-InsB融合蛋白对延缓NOD小鼠Ⅰ型糖尿病发作的研究
Oral administration of CTB insulin B chain produced in silkworm delays the diabetes development in NOD mice
【摘要】 为对霍乱毒素B亚基(the cholera toxin B subunit,CTB)联合胰岛素B链(insulin Bchain)表达的融合蛋白CTB-InsB长期口服延缓糖尿病发作的效果进行研究,将Ⅰ型糖尿病动物模型NOD小鼠随机分组,给予口服融合蛋白CTB-InsB后,检测尿糖、血糖,观测CTB-InsB对模型小鼠发病率的影响.试验表明与口服野生蚕血的对照组相比,每次口服融合蛋白CTB-InsB 50μg对NOD小鼠糖尿病的发作具有明显延缓作用(P<0.05),而100μg和10μg的剂量虽有下降趋势但是却差异不显著(P>0.05).
【Abstract】 An fusion protein that insulin B-chain peptide(InsB)was conjugated with the cholera toxin B subunit(CTB) was administered orally to NOD mice which could develop type Ⅰ diabetes spontaneously,in order to demonstrate the diabetes delay effect of CTB-InsB.The urine glucose then was observed,the blood glucose was detected and the effect of the fusion protein was analyzed.Compared with the control group which was fed with wild type silkworm blood lymph,this fusion protein at the dosage of 50 μg per feeding can statistically efficaciously prevents the mice from the rapid development of diabetes(P<0.05).Whereas the administration of 100 μg and 10 μg per feeding appears not so distinctly to decrease the incidence of diabetes (P>0.05),although those dosages still show downtrend.
