小球藻紫外线诱变及高产藻株筛选
The UV-Irradiation of Chlorella. vulgaris Beijerinck and Screening of Productive Strain
【摘要】 利用单细胞分离技术和紫外诱变育种技术,筛选高产小球藻株。通过单细胞分离技术获得的藻株Chlorella vul-garisQ7,生长速度和蛋白含量均优于其它藻株。以C.vulgarisQ7为出发株,进行紫外诱变,从120株诱变单克隆中筛选出3个株系:M51,M59,M73。与出发株相比,突变株M51,M59,M73的生长速率分别提高了6.23%,3.8%,5.92%。蛋白含量分别提高了2.5%,3.1%,1.9%。研究了诱变株的最适生长条件,结果表明在140μmol.m-2.s-1,25℃时,诱变株具有最大的生长速率和生物量。探讨了诱变株的遗传稳定性,结果表明诱变株可稳定遗传。
【Abstract】 Productive strains of Chlorella.vulgaris Beijerinck were filtrated by single cell clone combined with ultraviolet-irradition.Chlorella.vulgaris Q7,which grows faster and contains more protein than the others,was selected by single cell clone.Then the C.vulgaris Q7 was irradiated by ultraviolet to obtain mutants.We obtained 3 strains of mutants,named M51,M59 and M73,from 120 clones.Compared with the parent strain,the growth rates of mutants M51,M59 and M73 were increased by 6.23,3.8 and 5.92%,respectively and their protein contents were increased by 2.5,3.1 and 1.9%,respectively.The most suitable foster situation of productive strains was studied.It showed that the productive strains have the greater growth rate and bio-mass at 140 μmol·m-2·s-1,25 ℃.Their genetic stability was analysed also.Good genetic stability was concluded.
