酸性蛋白酶生产菌种的选育及培养基的优化
Breeding of Acid Protease-producing Strains & Optimization of Its Culture Medium
【摘要】 以黑曲霉(Aspergillus niger)为出发菌株,经原生质体紫外诱变,得到了产酸性蛋白酶优良菌株。采用单因素和正交实验,对其液态发酵的培养基成分进行了优化,得到最佳培养基组成为:玉米粉1%,豆饼粉3%,鱼粉1.2%,氯化铵4%,磷酸氢二钾0.3%,氯化钙0.5%。在优化的培养基条件下,30℃、180 r/min摇瓶发酵72 h,酶活力达到1117.52 u/mL。
【Abstract】 Quality acid protease-producing strains were obtained as follows: aspergillus niger used as starting strains and then it underwent protoplast ultraviolet mutation.The culture medium compositions for its liquid fermentation were optimized through single factor experiment and orthogonal test as follows:1 % corn powder,3 % bean cake powder,4 % ammonium chloride,0.3 % dipotassium hydrogen phosphate,and 0.5 % calcium chloride.The enzyme activity of the strains could reach 1117.52 u/mL through 72 h flask-shaking(180 r/min) fermentation at 30 ℃.
