CXC趋化因子配体10联合顺铂抑制LL/2肿瘤肺转移的实验研究

【摘要】 目的研究CXC趋化因子配体10(CXCL10)联合顺铂对荷瘤小鼠LL/2肿瘤肺转移的治疗效果。方法小鼠Lewis肺癌LL/2肿瘤模型建立在C57BL/6小鼠上。实验小鼠随机分4组:CXCL10+顺铂组;CXCL10组;顺铂组;PBS对照组。CXCL10按25μg/(kg·d)使用,皮下注射,连用30d。顺铂采用腹腔内注射[5mg/(kg·week)],于CXCL10治疗的第14、21天使用,连用2个周期。观察肿瘤体积与小鼠生存时间,观察各组小鼠肺表面的肿瘤转移结节数。结果CXCL10治疗明显抑制了Lewis肺癌的肺转移,肺表面的肿瘤结节数少于对照组(P<0.05)。联合治疗组表现较强的抗肿瘤疗效,其肿瘤生长速度减慢,治疗效果(P<0.01)优于顺铂或者CXCL10组(P<0.05)。结论CXC趋化因子配体10联合顺铂更有效地抑制LL/2肿瘤肺转移和延长生存期。

【Abstract】 Objective To study the inhibition of pulmonary metastasis in LL/2 tumor-bearing mice by CXC chemokine ligand 10 (CXC-L10) combined with cisplatin. Methods Lewis lung cancer (LL/2) models were established in the C57BL/6 mice. Animals were subdivided into four groups and treated with PBS, CXC-L10, cisplatin or CXCL10 plus cisplatin, respectively. Treatment were performed with either CXCL10 s.c. at 25 g/(kg·d) once a day for 30 days, cisplatin cycled twice (5 mg/kg i.p. on days 14 and 21 after the initiation of CXCL10), or both agents together. Tumor volume and survival time were observed. Lung metastasis nodules were further counted under dissecting microscope at day 4 after terminating treatment. Results Metastatic nodules in CXCL10 treated group were decreased compared with PBS control (P<0.05). CXCL10 plus cisplatin reduced tumor growth more effectively (P<0.01), although cisplatin or CXCL10 individually resulted in suppression of tumor growth (P<0.05). Conclusion CXCL10 combined with cisplatin further inhibited the pulmonary metastasis of LL/2 tumor and prolonged the survival time of tumor bearing-mice.
