牛脊髓前角匀浆免疫后豚鼠血清和脑脊液氨基酸的测定
Study on contents of serum and CSF amino acids in the Guinea pig immunized by bovine spinal cord ventral horn homogenate
【摘要】 目的:牛脊髓前角匀浆免疫豚鼠导致免疫介导的运动神经元损伤,通过测定血清与脑脊液中谷氨酸、天冬氨酸、甘氨酸及γ-氨基丁酸含量探讨其发病机制。方法:1.用新鲜牛脊髓前角匀浆免疫豚鼠,观察豚鼠肢体活动情况,进行神经功能评估、电生理检查、后肢肌肉活检及固定液灌注后取颈、腰膨大。观察运动神经元变性情况。2.应用HPLC检测血清和脑脊液谷氨酸、天冬氨酸、甘氨酸及γ-氨基丁酸含量。结果:1.免疫组豚鼠出现肢体瘫痪,体重减轻,肌电图示失神经支配,肌肉活检有束样萎缩。颈、腰膨大前角运动神经元变性,数量减少。2.瘫痪豚鼠血清和脑脊液谷氨酸、天冬氨酸升高,甘氨酸和γ-氨基丁酸无变化。结论:氨基酸的兴奋毒作用参与了免疫介导的运动神经元损伤过程。
【Abstract】 Objective:Using bovine spinal cord anterior horn homogenate immuned guinea pigs to induce immune mediated motor neuron damage,then investigate the changes of serum and CSF glutamic acid,glycine andγ-aminobutyric acid level using HPLC to study the aetiology.Methods:Guinea pigs were immunized with fresh bovine spinal cord ventral horn homogenate and were examined for neurological evaluation every day.EMG examincotion was given every four week. The hind limb muscle was used for histological examination. Animals were perfused with a solution of 4%paraformaldehyde,0.05%glutaraldehyde and 0.2% picric acid.Spinal cord was post-fixed by immersion in the same fixative for 3 days.Cervical and lumbar enlargements were used for pathological examination.Levels of serum and CSF amino acids were measured via high performance liquid chromatography.Results:Guinea pigs immunized with bovine spinal ventral horn homogenate develop muscle weakness with electromyographic evidence of denervation. Muscle biopsy found atrophic fibers in group.Pathological examination demonstrates loss and degeneration of motoneuron in the spinal cord.The level of glutamic acid and aspartic acid was increased in the paralyzed animals’serum and CSF,glycine and GABA had no significant change.Conclusions:Excitatory amino acids play a role in the immune induced motor neuron damage process.
