彗星试验检测亚慢性吸入二硫化碳对小鼠白细胞DNA的损伤情况

【摘要】 [目的]检测亚慢性吸入二硫化碳对小鼠白细胞DNA的损伤情况。[方法]以浓度为0、400、800、1200 mg/m~3的二硫化碳蒸气给小鼠做静式吸入染毒5周,每天2h。用单细胞凝胶电泳实验(彗星试验)检测二硫化碳对白细胞DNA的损伤。[结果]低、中、高染毒组与对照组小鼠白细胞彗星的拖尾率分别为68.40%、86.20%、97.40%和4.60%,经X~2检验各试验组与对照组差异有统计学意义(P<0.01);各试验组彗星头部百分比分别为(61.13±5.54)%、(46.15±8.37)%、(32.53±10.21)%,与对照组彗星头部百分比(90.65±3.74)%差异有统计学意义(P<0.001);尾长分别为(12.12±2.55)、(26.19±5.19)、(39.83±9.21)μm,与对照组(1.29±0.47)μm差异均有统计学意义(P<0.001)。染毒剂量与彗星头部百分比的相关系数为-0.981,与尾长的相关系数为0.998,P值均<0.001。[结论]亚慢性吸入二硫化碳可损伤小鼠白细胞DNA,且随着染毒剂量的增加,损伤程度也越严重,存在剂量-反应(效应)关系。

【Abstract】 [Objective]To detect the DNA damage of sub-chronic inhalation of carbon disulfide on white blood cells (WBC)of mice.[Methods]The damage of WBC of mice exposed to carbon disulfide by 2 h static inhalation per day at different concentrations(400,800,1600,and 0mg/m~3)for 5 weeks was analyzed with Single Cell Gel Electropboresis(SCGE or Comet Assay).[Results]In the four groups,the rates of damage were 68.40%,86.20%,97.40% and 4.60%,respectively.There were significant differences between experiment groups and the control(P<0.01).The percentages of head of the comet were (61.13±5.54)%,(46.15±8.37)%,(32.53±10.21)% and(90.65±3.74)% respectively,also significantly different among experiment groups and the control(P<0.001),r=-0.981.The tail lengths in the 4 groups were(12.12±2.55),(26.19±5.19), (39.83±9.21)and(1.29±0.47)μm respectively,with significant differences between each experiment group and the control(P<0.001),r=0.998.[Conclusion]The sub-chronic carbon disulfide exposure could induce DNA damage.As the increase of dose, the DNA damage of WBC got more severe,which lead to a dose-response(effect)relationship.
