预测急性冠状动脉综合征危险因素的候选基因多态性
Prediction of the risk of acute coronary syndrome from polymorphisms in candidate genes
【摘要】 目的:探讨中国汉族急性冠状动脉综合征(ACS)患者的易感性基因。方法:采用聚合酶链式反应/限制性片段长度多态性分析中国汉族108例患者(ACS组)和90例健康者(对照组)的4个候选基因,7个多态性。结果:ACS组基质金属蛋白酶-2(MMP-2)(-1306C/T)C等位基因频率与对照组比较差异有统计学意义(P<0.05),2组间3种基因型(CC、CT和TT)分布差异有统计学意义(P<0.05),而ACS组MMP-2(-1575G/A,-790G/T和-735C/T)、基质素-1(5A6A)、G蛋白β3亚单位(825C/T)和连接蛋白37(CX37)基因多态性(1019C/T)基因型分布和等位基因频率与对照组比较差异无统计学意义(P>0.05)。结论:MMP-2基因(-1306C/T)多态性可能与中国汉族人群ACS有关,MMP-2基因-1306C等位基因可能是ACS遗传易感性的基因标志之一。
【Abstract】 Objective:To explore the susceptibility gene in Chinese Han Nationality with acute coronary artery syndrome(ACS).Method:Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)method was used to determine the genotypes of 7 polymorphisms of 4 candidate genes in 108 patients with ACS and 90 healthy control subjects from Chinese Han nationality.Result:The frequenclgs of MMP-2(-1306C/T)polymorphism C allele(50.5%∶40.9%)was higher in the ACS group than that in healthy control(P<0.05).The frequencies of three genotypes(CC,CT and TT)were significantly different between the two groups(P<0.05),whereas the genotypes and allele frequencies of MMP-2(-1575G/A,-790G/T and-735C/T),Stromelysin-1(5A6A),G Protein β3 subunit(825C/T)and Connexin 37(1019C/T)gene polymorphism in ACS group were not significantly different from those controls(P>0.05).Conclusion:The present study suggests that the genetic polymorphism in MMP-2 gene(-1306C/T)promoter is associated with the susceptibility to ACS in Chinese Han nationality.
