抑癌基因在膀胱癌组织中的表达及其与Akt磷酸化的相关性

【摘要】 目的:探讨抑癌基因PTEN在膀胱移行细胞癌(简称膀胱癌)的表达及其与蛋白激酶B(PKB/AKT)磷酸化的相关性。方法:采用免疫组织化学S-P染色法,Western印迹杂交法检测46例膀胱癌组织中PTEN的表达,并与13例腺性膀胱炎、10例正常膀胱组织进行对照;同时分析膀胱癌组织中PTEN表达与AKT磷酸化水平的关系。结果:膀胱癌组织中PTEN蛋白的阳性表达(60.9%)明显低于腺性膀胱炎(95.4%)和正常膀胱组织(100%)(P<0.01);同时低分化、有肿瘤转移的膀胱癌的PTEN表达强度明显低于高分化、无转移的膀胱癌(P<0.05);膀胱癌组织中PTEN蛋白表达和Akt磷酸化水平呈明显负相关(P<0.01)。结论:由于PTEN蛋白表达下调或缺失,导致AKT磷酸化水平的明显增高,在膀胱癌的发生、发展中起着重要的作用。

【Abstract】 Objective:To investigate the clinical significance of phosphatase and tensin homolog deleted on chromosome ten(PTEN)protein exppression and it’s correlation with phosphorylation of protein kinase B(PKB/Akt)in human bladder transitional cell Carcinoma(BTCC). Methods:The expression of PTEN and phospho-Akt were detected by SP immunohistochemical technique and Western blotting in 46 cases of BTCC, 13 cases of cystitis glandular and 10 cases of normal bladder tissues, respectively. Results:The positive expression of PTEN in BTCC(60.9%, 0.083±0.018) were significantly lower than those in cystitis glandular and normal bladder tissues(P<0.01). The expression level of PTEN was related to the differentiation degree of BTCC and the status of metastasis(P<0.05). Western blotting revealed a significant inverse correlation between PTEN and phospho-Akt(r=-0.786,n=46,P<0.01).Conclusions:These results indicate that down regulation of PTEN, which may not be able to effectively inhibit the hyperphosphorylation of PKB/Akt, will be an important step in oncogenesis and progression of BTCC.
