秀丽线虫sod-3基因RNA干扰及早衰模型的建立
Model of Rapid Aging Based on RNAi-Mediated Gene Silencing of sod-3 Expression in Caenorhabditis Elegans
【摘要】 利用RNA i技术,通过干扰秀丽线虫中的sod-3基因表达,建立了因超氧化物歧化酶3(SOD3)表达缺失引起快速衰老的线虫模型,为sod-3基因功能的研究以及相关疾病的基因治疗等提供动物模型.
【Abstract】 According to the oxidative damage theory of aging,free radicals,particularly reactive oxygen species(ROS),are responsible for the senescence and many human diseases.ROS are generated during normal metabolic process in all the oxygen-utilizing organisms.They are detoxified by superoxide dismutase.The manganese-containing superoxide dimutase 3(SOD3) has been shown to play a role in the aging process in Caenorhabditis elegans.In this research,we show that sod-3 RNA interference(RNAi) induces insenecence in Caenorhabditis elegans as expected.This method can be used to understand the process of aging and the mechanism of antioxidance.
