川穹嗪对血管内皮细胞损伤的保护作用

【摘要】 目的:观察川穹嗪对氧化型低密度脂蛋白(ox-LDL)致人内皮细胞ECV304损伤的保护作用及机制,探讨其在动脉粥样硬化防治中的意义。方法:体外培养内皮细胞ECV304,分为4组:空白对照组(ECV304)、川穹嗪对照组(ECV304+川穹嗪)、ox-LDL组(ECV304+ox-LDL)和川穹嗪保护组(ECV304+ox-LDL+川穹嗪)。各组中ox-LDL、川穹嗪终浓度分别为0.1、0.5mg/ml。细胞加入相应处理因素后,孵育24h,收集上清液,测定乳酸脱氢酶(LDH)、丙二醛(MDA)含量;收集细胞检测超氧化物歧化酶(SOD)、谷光甘肽过氧化物酶(GSH-PX)活性。结果:川穹嗪保护组LDH含量为(75.34±6.89)U/L,MDA为(0.64±0.08)μmol/L,而ox-LDL组分别为(468.34±9.46)U/L、(1.58±0.02)μmol/L,川穹嗪保护组LDH、MDA含量均明显低于ox-LDL组,差异有显著性(P<0.05);同时川穹嗪保护组SOD、GSH-PX活性明显高于ox-LDL组,川穹嗪保护组SOD为(40.43±6.52)mU/L,GSH-PX为(125.63±4.67)mU/L,ox-LDL组SOD为(27.43±5.43)mU/L,GSH-PX为(74.43±6.39)mU/L,2组差异有显著性(P<0.05)。结论:川穹嗪对ox-LDL导致內皮细胞损伤具有保护作用,机制可能与其抗氧化效应有关。

【Abstract】 Objective:To observe the effect of Tetramethylprazine (Tet) on endothelial cell(ECV304) injured by oxidative low-density lipoprotein (ox-LDL).Methods:Cultured ECV304 cells were divided into 4 groups: Control group (ECV304),Tet control group (ECV304+ Tetramethylprazine),ox-LDL group (ECV304+ox-LDL) and Tet group (ECV304+ox-LDL+Tetramethylprazine).The concentrate of ox-LDL and Tet used in the experiment was 0.1 mg/ml and 0.5 mg/ml respectively.After dealing with different treatments,cells of four groups cultured for further 24 hours,and then culture medium was collected for the measurement of LDL,MDA contents.The activities of SOD and GSH-PX were detected in ECV304 cells.Result:The contents of LDH and MDA in Tet group were significantly lower than ox-LDL group (P<0.05);The activities of SOD and GSH-PX in Tet group were significantly increased than ox-LDL group (P<0.05).Conclusion: Tetramethylprazine has the protective function of the endothelial cells injury induced by ox-LDL via a anti-oxidation effect.
