三七总皂甙对大鼠脊髓横断损伤后NGF和BDNF表达的影响

【摘要】 目的探讨三七总皂甙对大鼠脊髓全横断后NGF和BDNF表达的影响,阐明PNS治疗SCI的分子机制。方法40只成年健康SD大鼠,随机均分为假手术对照组(SO组),单纯脊髓横断组(tSCI组),生理盐水组(NS组),三七总皂甙组(PNS组)。每组于术后1天、7天各处死5只取材,观测形态结构和神经元数量,并采用免疫组织化学ABC法检测各组神经元细胞神经生长因子(NGF)和脑源性神经营养因子(BDNF)的表达。结果在假手术组NGF和BDNF的表达较少,脊髓腹角神经元数量、NGF和BDNF的表达无时间变化趋势;tSCI后,脊髓腹角神经元数量明显减少,NGF和BDNF表达增加,损伤后时间越长,神经元数量减少越明显,NGF和BDNF阳性的细胞数增加越多;PNS组脊髓腹角神经元数量较tSCI组、NS组多(P<0.05),但较SO组少(P<0.05),NGF和BDNF的表达较SO组t、SCI组和NS组增强(P<0.05)。结论PNS可使tSCI后神经元数量增加,NGF、BDNF表达增加,表达时间提前,PNS对SCI有保护和治疗作用,其作用机制可能NGF、BDNF表达增强有关。

【Abstract】 Objective To observe the effect of panax notoginseng saponins(PNS) on the expression of NGF and BDNF in transected spinal cord of rat and explore the therapeutic mechanism of PNS for SCI. Methods 40 adult SD rats were randomly divided into 4 groups: SO group,tSCI group,NS group and PNS therapy group.Five rats in each point were sacrificed at 24h or 7d after operation.The immunohistochemical ABC method was used to detect the expression of NGF and BDNF in each group and observe the structure and the number of survival neurons.Results The number of neurons in the ventral horn of spinal cord in tSCI group had no apparent change compared with that of NS group at 24 h and 7d,but that of PNS group increased more significantly than that of tSCI group and NS group(P<0.05).And the number of neurons in SO group was significantly higher than that of the above three groups at any time(P<0.05).The increasing expression of NGF and BDNF positive cells was observed at 24 h after operation in PNS group and reach the peak value in 7ds after operation,and that of PNS group were significantly higher than that of tSCIgroup and NS group(P<0.05).The expression of the NGF and BDNF was different in PNS group,tSCI group and NS group at 24h and 7d. And there is no difference between tSCI group and NS group at 24h and 7d(P>0.05).The expression of NGF and BDNF in SO group were significantly lower than that of the above three groups at any time(P<0.05).Conclusions PNS can significantly enhance the expression of endogenous NGF and BDNF,and also maintain the survival number of the neurons after tSCI of the rats.The mechanism of protection and therapy of PNS may be related to the increasing expression of endogenous NGF and BDNF.
