康普瑞丁磷酸二钠在体内外的抗肿瘤作用
Antitumor effects of Combretaftation A-4P in vivo and in vitro
【摘要】 目的观察化合物康普瑞丁磷酸二钠(CA-4P)对人早幼粒细胞白血病HL-60细胞的体外抑制作用及对裸小鼠腹腔移植HL-60细胞的体内抗肿瘤作用。方法分别用MTT法、细胞生长曲线及细胞集落形成试验,研究CA-4P对HL-60细胞增殖的抑制作用,台盼蓝染色观察细胞形态变化;HL-60细胞腹腔移植裸小鼠,iv给予CA-4P,qd,5 d,观察裸小鼠的腹水形成情况及生存时间,计算生命延长率。结果MTT法提示,0.5~64.0μg.ml-1CA-4P对HL-60细胞48 h的抑制率为15%~79%,平均IC50约为6.579μg.ml-1;生长曲线提示,CA-4P对HL-60细胞的抑制作用呈明显时效和量效关系;台盼蓝染色后细胞出现凋亡形态的变化。CA-4P对裸小鼠腹腔移植HL-60细胞的治疗作用呈量效关系,随剂量增大,裸小鼠的生命延长率也增加。结论CA-4P对人早幼粒细胞白血病HL-60细胞的增殖具有抑制作用,对HL-60细胞裸小鼠腹腔移植瘤具治疗作用。
【Abstract】 OBJECTIVE To investigate the in vivo/in vitro inhibitory effects of compound Combretaftation A-4P(CA-4P) on human leukemia HL-60 cell line and on HL-60 cell inoculated with transplant tumor nude mice.METHODS The inhibitory effect of compound CA-4P on human leukemia cell line HL-60 was assayed by MTT analysis,cell growth curve and clone forming experiments with different concentrations of CA-4P.The change of cell configuration was observed by dyeing.The inhibitory effect of compound CA-4P on nude mice inoculated with HL-60 cells in vivo was demonstrated by the prolongation of survival times in mice carried tumor.RESULTS After treated with CA-4P for 48 hours,MTT assay showed the dose range from 0.5 μg·ml-1 to 64.0 μg·ml-1 had the inhibitory ratio range from 15% to 79%.The average IC50 in 48 h was 6.579 μg·ml-1.The dose-effect relationship was described in the growth curve.The characteristic typical morphology for apoptosis was observed under microscope.The dose-effect relationship was also observed in the experimentation of HL-60 cell inoculated transplant tumor nude mice by CA-4P, and the prolongation of survival times in mice carried tumor was prolonged along with the rise of CA-4P concentration.CONCLUSION CA-4P could inhibit the growth of HL-60 cell in vitro,and its in vivo inhibitory effectin of HL-60 cell inoculated nude mice was distinct.
