超淬灭分子信标的合成与性能研究
The synthesis and investigation of super-quencher molecular beacon
【摘要】 合成并纯化了普通DNA引物、常规分子信标和超淬灭分子信标,利用基质辅助激光解析电离飞行时间质谱对产物进行解析,证明了其为目标产物。与cDNA杂交的结果表明,超淬灭分子信标的信背比达到40倍,比常规分子信标高出4倍,有望实现对样品的高灵敏检测。
【Abstract】 Three kinds of nucleic acids probes were synthesized and purified,which included cDNA,normal molecular beacon(MB),super-quencher molecular beacon(SQ-MB).The products were identified by MALDI-TOF-MS.The hybridization result of SQ-MB with cDNA showed that the signal/background ratio increased 40 times,and it was 4 times than normal molecular beacon.The super-quencher molecular beacon with excellent sensitivity has the potential for high sensitive detection of target nucleic acids.
