【摘要】 目的观察大鼠脑出血后海马线粒体DNA含量改变及不同中医治法的干预作用,探讨中医最佳治法及方药。方法用Ⅶ型胶原酶于脑内定位注射诱导大鼠脑出血模型,紫外分光光度计测定脑海马mtDNA水平。结果145只大鼠全部进入结果分析:①mtDNA水平模型组和中药干预组各时间点均显著低于假手术组(P<0.01),假手术组与正常对照组比较差异无统计学意义(P>0.05),采用不同中医治法后,在第1天平肝熄风汤与大承气汤组的mtDNA含量较模型组增高差异具有统计学意义(P<0.01),在第3、7天,风、火、痰、瘀同治与分治各组的mtDNA含量均较模型组显著增高差异具有统计学意义(P<0.01),而以风、火、痰、瘀同治(平肝熄风汤)组mtD-NA又明显高于风、火、痰、瘀分治各组差异具有统计学意义(P<0.01)。②神经功能缺损积分:模型组和各中药干预组大鼠于造模后4 h神经功能缺损积分达高峰,随后逐渐下降,至第7天仍高于假手术组和正常组差异具有统计学意义(P<0.01),采用不同中医治法治疗后于第1、3、7天其神经功能缺损积分均低于模型组差异具有统计学意义(P<0.05),于第3、7天平肝熄风汤组积分又比风、火、痰、瘀分治各组明显降低差异具有统计学意义(P<0.05)。结论大鼠脑出血后海马线粒体内DNA含量下降,而采用不同中医治法均能阻止mtDNA水平下降,改善神经功能缺损,但以风火痰瘀同治之平肝熄风汤效果最为明显。更多还原

【Abstract】 Objective To determine the change of mitochondrial DNA(mtDNA) content after intracerebral hemorrhage(ICH) in rats and the interventional effects of different treatments in TCM and investigate the best therapy and prescription.Methods The experimental ICH models are induced by Type Ⅶ collagenase stereotaxically injected into the globas pallidus.And the Level of hippocampal mtDNA was detected with ultraviolet spectrophotometer.Results In all 145 rats,①the mtDNA level in model group and different TCM treatments groups was lower than that in sham operation group at each time points(P<0.01),but without significance between sham operation group and normal control group(P>0.05).The mtDNA level in Pinggan xifeng decoction group was higher than that in model group and other treatment groups at 3 th and 7th days after modeling(P<0.01).②Scores of neurological defects in model group and different interventional groups with Chinese drugs reached the peak 4 hours after modeling,then decreased gradually,but still were more than those in sham operation group and normal group at 7th day(P<0.01),and the equation scores of Pinggan xifeng decoction group at 3th and 7th day was decreased significantly than other treatment groups with wind,fire,sputum and blood stasis syndromes respectively(P<0.01).Conclusion Hippocampal mtDNA content decreases in rats with ICH,but the mtDNA level can be blocked if Chinese drugs therapies,which is effective to improve neurological impairment,can be used,and Pinggan xifeng decoction is the best effective for ICH.更多还原
