【摘要】 【目的】研究趋化因子受体CXCR4对不同转移潜能骨肉瘤细胞株的裸鼠肺转移的影响。【方法】采用单细胞克隆技术,从人骨肉瘤细胞系MG-63中分离培养出2株不同CXCR4表达的骨肉瘤细胞株,免疫组化检测其CXCR4的表达水平,将骨肉瘤细胞株分别接种于裸鼠的颈背部皮下并于4周后处死裸鼠,观察肺部转移情况。【结果】分离培养出2株骨肉瘤细胞株,其CXCR4蛋白的表达水平为B8>G5,各组裸鼠的肺组织转移灶数为B8>G5,与CXCR4蛋白的表达呈正相关。【结论】骨肉瘤细胞高度的肺转移能力与CXCR4的高表达密切相关。更多还原

【Abstract】 【Objective】To study effects of chemokine receptor CXCR4 expression on pulmonary metastasis of osteosarcoma cell sublines in nude mice.【Methods】Osteosarcoma cell sublines were obtained by the unicell cloning technique from human osteosarcoma cell line MG-63,and the expression of protein of CXCR4 was detected by immunohistochemistry.Then osteosarcoma cell sublines with different CXCR4 expression were injected into cervical and dorsal subcutaneous tissue of nude mice,which were killed after 4 weeks.Pulmonary metastasis of different cell sublines was observed.Serial section of the fixed lung tissue was done to count and grade.【Results】 Two osteosarcoma cell sublines(B8,G5) were got from MG-63.And there was a significant difference in the expression of protein of CXCR4 between them(B8>G5).Superficial and interior pulmonary metastases in nude mice were also different(B8>G5),and had positive correlation with the expression of CXCR4.【Conclusion】 The potency of high endosomatic pulmonary metastasis of human osteosarcoma cells is intimately associated with high expression of CXCR4.更多还原
