【摘要】 目的:探讨雷公藤内酯醇(Triptolide,Tri)抑制体外培养的牛晶状体上皮细胞(bovine lens epi-thelial cell,BLECs)增殖及其细胞内cGMP浓度变化的机制。方法:取第4代BLECs,分3组培养。空白对照组:以DMEM培养;增殖对照组:以DMEM+50μg/L rhEGF培养;雷公藤内酯醇组:增殖对照组+20μg/L雷公藤内酯醇。用MTT比色法检测增殖抑制率来测定Tri 20μg/L对细胞增殖的影响,用放射免疫法测定BLECs内cGMP浓度。结果:浓度为20μg/L的Tri、作用在处于增殖状态的BLECs时间分别为6,12,24,48,72 h,均出现明显的增殖抑制作用,其增殖抑制率升高呈现明显的时间依赖性。而雷公藤内酯醇组细胞内cGMP含量比增殖对照组明显降低。结论:Tri对晶状体上皮细胞增殖有明显的抑制作用,而该作用可能通过降低细胞内cGMP浓度来完成。更多还原

【Abstract】 Objective: To investigate the effects of Triptolide on the proliferative inhibition and intracellular cGMP in cultured bovine-lens epithelial cells(BLECs) in vitro.Methods:The fourth generation of BLECs were collected and cultured into 3 groups : the first group with DMEM,the second with DMEM +50 μg/L rhEGF and the third with 50 μg/L rhEGF+20 μg/L Tri +DMEM. The effects of Triptolide on the proliferation BLECs were measured with MTT colorimetry and the intracellular cGMP was measured by radioimmunoassay.Results: Triptolide at 20 μg/L concentration showed significant inhibitory effect on rhEGF-induced proliferation of BLECs at 6 h,12 h,24 h,48 h and 72 h time courses respectively,the ratio of proliferative inhibition enhancl distinctively on time dependence.Intracellular cGMP concentration of BLECs cultured with 50 μg/L rhEGF+20 μg/L Tri +DMEM was lower than that of the group cultured with DMEM +50 μg/L rhEGF.Conclusion:Triptolide could effectively inhibit the proliferation of BLECs in vitro.The inhibitory effects of triptolide on the proliferation may accomplished by changing the concentration of the cytosolic cGMP.更多还原
