罗格列酮对环孢素A作用下大鼠肾脏成纤维细胞转化生长因子-β1和层黏连蛋白表达的影响

【摘要】 目的:观察罗格列酮(RGZ)对环孢素A作用下大鼠肾脏成纤维细胞(NRK)转化生长因子-β1(TGF-β1)和层黏连蛋白(FN)表达的影响,探讨罗格列酮对环孢素A肾毒性的改善作用及其机制。方法:体外培养NRK细胞,在无血清DMEM低糖培养基同步化24h后,随机分为7组:正常对照组,CsA0.25mg/L组,CsA0.5mg/L组,CsA1.0mg/L组,CsA2.0mg/L组,CsA1.0mg/L加RGZ10μmol/L组,CsA1.0mg/L加RGZ15μmol/L组,继续培养24h后,收集细胞,用逆转录-聚合酶链反应(RT-PCR)方法检测各组细胞中TGF-β1mRNA表达,用蛋白印迹(Western blotting)方法检测FN蛋白的表达。结果:不同质量浓度的CsA均可促进TGF-β1 mRNA和FN蛋白表达(P<0.05),罗格列酮能显著下调CsA引起的TGF-β1 mRNA和FN蛋白的高表达。结论:罗格列酮可通过下调CsA引起的TGF-β1和FN的高表达,抑制CsA导致的肾脏成纤维细胞外基质的蓄积。

【Abstract】 Aim:To investigate the protective effects of rosiglitazone (RGZ) on rat renal interstitial fibroblasts (NRK) damaged by cyclosporine A(CsA).Methods:The NRK cells were cultured in vitro and divided into seven groups: A:normal group, B:CsA 0.25 mg/L group,C:CsA 0.5 mg/L group,D:CsA 1.0 mg/L group,E:CsA 2.0 mg/L group,F:CsA 1.0 mg/L + RGZ 10 μmol/L group,G:CsA 1.0 mg/L + RGZ 15 μmol/L group. After 24 h the mRNA expression of TGF-β1 was examined by RT-PCR,and the protein expression of fibronectin (FN) was examined by Western blotting.Results: Compared with normal group,CsA stimulated the expression of TGF-β1 and FN in different concentration, and RGZ inhibited those changes significantly.Conclusion: RGZ can reduce the high expression of TGF-β1 and FN induced by CsA, and inhibit the cumulation of extracellular matrix in kidney fibroblasts.
