转基因抗穗发芽小麦的获得
The study on transgenic wheat with the characteristic of pre-harvest sprouting resistance
【摘要】 以易穗发芽小麦幼胚为受体,用基因枪法对TrxS反义基因(目的基因)和Bar基因(标记基因)进行了共转化;通过对轰击后的小麦幼胚进行愈伤组织诱导、胚性愈伤组织分化和生根培养,获得再生植株;经分子检测证实在3个受体材料上获得转基因再生植株和可遗传的转基因后代.通过培养皿籽粒发芽试验、离体整穗保湿发芽试验和温室模拟降雨穗发芽试验,均证明转基因后代材料具有延缓籽粒发芽和抗穗发芽的特性.
【Abstract】 Wheat(Triticum aestivum L) pre-harvest sprouting,a world problem,effects the grain quality,yield and economic value.In the study,the easily pre-harvest sprouting,white seed varieties were selected as receptor materials.Antisense TrxS gene and Bar gene were co-transformed to immature embryos of wheat via microprojectile-bombardment.The bombarded immature embryos were cultured for inducing callus,callus differentiation and plant regeneration and the regenerated plants were obtained.The molecular assay of the regenerated plants showed that the transgenic plants and their progenies were achieved from 3 receptor genotypes by PCR,nest-PCR,PCR-southern and Southern blotting.The seed germination and pre-harvest sprouting tests indicated that the transgenic wheat lines have possessed the strong anti-sprouting characteristics.
