三系不育系水稻叶绿体DNA遗传多态性研究
Studies on chloroplast DNA polymorphism of CMS line rice
【摘要】 对11个三系不育系水稻、2个典型籼稻、2个典型粳稻和3个普通野生稻叶绿体DNA的ORF100,ORF29-TrnCGCA,rps16基因内含子和TrnTUGU-TrnLUAA转录间区4个片段进行扩增,并对后两个片段测序.研究结果表明,三系不育系水稻叶绿体DNA的ORF100和ORF29-TrnCGCA两片段的长度均与典型的籼稻一致,即供试的11个三系不育系水稻都为籼型叶绿体.11个三系不育系材料中有10个材料在测序的rps16基因内含子TrnTUGU-TrnLUAA转录间区的序列完全一致,红莲型不育系粤泰A与其他10种不育系水稻在rps16片段上有一个单碱基和一个6碱基片段的差异.由此说明,与供试的典型籼稻、典型粳稻和普通野生稻相比,三系不育系细胞质的多态性水平非常低.
【Abstract】 We amplified ORF100、ORF29-TrnCGCA、intron of rps16 gene and transcribed spacer of TrnTUGU-TrnLUAA,and sequenced the later two segments of 11 CMS line rice 、2 typical indica、 3 typical japonica and 3 common wild rice.The result showed the length of ORF100 and ORF29-TrnCGCA was consistent of typical indica had,that said 11 CMS line rice had indica chloroplast.The 10 CMS line rice of transcribed spacer of TrnTUGU-TrnLUAA had completely same sequence,the Yue-tai A of HL-cytoplasmic male sterility rice had difference of 1 bp base and 6 bp base.From these,the CMS line rice had far lower polymorphism of cytoplasmic than typical indica、typical japonica and common wild rice.
