硒和锗对氟染毒大鼠血清和组织中钙和镁及锌的影响

【摘要】 目的研究硒锗联合作用对氟中毒大鼠体内钙、镁、锌含量的影响。方法将健康初断乳清洁级SD大鼠随机分为对照组、氟暴露组、氟+硒暴露组、氟+锗暴露组、氟+硒+锗暴露组,每组10只。除对照组可自由饮用蒸馏水外,其余各组大鼠均自由饮用100mg/L的NaF溶液。氟+硒暴露组、氟+锗暴露组、氟+硒+锗暴露组分别采用20mg/L的Na2SeO3、2000mg/L的Ge-132、20mg/L的Na2SeO3和2000mg/L的Ge-132,按5ml/kg(0.5ml/100g)进行灌胃,共饲养90d。每周称体重1次,并观察大鼠的活动、饮食和一般状况。采用火焰原子化法测定大鼠血清和肝、肾组织中钙、镁、锌的含量,用离子色谱法测定大鼠血清和肝、肾组织中的氟含量。结果从第21天开始,氟暴露组大鼠体重增长明显慢于对照组,大鼠活动减少,皮毛不舒展,有脱毛现象;其余各组体重增长情况介于对照组、氟暴露组之间。氟暴露组大鼠血清、肝、肾组织中氟含量最高(P<0.05),对照组最低。与氟+硒+锗暴露组相比,氟+锗暴露组肝、肾组织和氟+硒组肾组织中氟含量较高,差异均有统计学意义(均P<0.05)。对照组、氟+硒暴露组、氟+锗暴露组、氟+硒+锗暴露组大鼠血清、肝、肾组织中钙、镁含量高于氟组,差异均有统计学意义(均P<0.05)。与氟+硒+锗暴露组相比,氟+锗组、氟+硒组和对照组肝、肾组织中钙含量以及氟+锗组、氟+硒组和对照组血清、肝组织中镁含量较低。氟+硒+锗组血清、肝、肾组织中锌含量高于对照组、氟+硒组、氟+锗组、氟组。结论硒锗联合对氟诱导的血清、肝、肾组织中钙、镁、锌含量降低具有明显的拮抗作用,且强于单独的硒或锗的拮抗作用。

【Abstract】 Objective To understand the combined effects of selenium (Se), germanium (Ge) on the level of Ca, Mg, Zn in the tissues of rats exposed to fluoride(F). Methods SD rats were divided into 5 groups and respectively treated with distilled water(control), 100 mg NaF/L(through drinking water), based on the fluoride treatment, Se, Ge were respectively given by gavage at the doses of 0.1 mg Na2SeO3/(kg·d), 10 mg Ge-132/(kg·d) and 0.1 mg Na2SeO3/(kg·d) plus 10 mg Ge-132/(kg·d) for 90 days. The body weight, activity, and general state were observed. The content of Ca,Mg,Zn in the serum,liver and kidneys were determined by flame atomization method. The content of F was detected by ion chromatography. Results The body weight in F+Se, F+Ge and F+Se+Ge groups were higher than that in F group and lower than that in the control group. The content of F in the liver, kidney in F+Se, F+Ge groups were higher than that in F+Se+Ge group(P<0.05). Ca, Mg content in serum, liver and kidney in Se, Ge groups were higher than that in F group(P<0.05). Ca, Zn content in the liver and kidney in F+Se+Ge were higher. Conclusion The combination of Se and Ge has a powerful antagonistic effects to fluoride toxicity.
