低剂量甲苯吸入诱导大鼠条件性位置偏爱的实验研究
Effect of Low Level Exposure of Toluene on CPP in Rats
【摘要】 目的研究低剂量甲苯吸入能否诱导大鼠条件性位置偏爱(CPP)及诱导CPP的途径。方法将60只SD大鼠随机分成低浓度甲苯染毒组(328.6mg/m3)、高浓度甲苯染毒组(1026.8mg/m3)、空白对照组、嗅球毁损模型组、嗅球毁损低浓度甲苯染毒组(328.6mg/m3)和嗅球毁损高浓度甲苯染毒组(1026.8mg/m3),每组10只。对染毒组进行甲苯亚慢性染毒4周(5d/周,6h/d),空白对照组及嗅球毁损模型组不予干预。染毒后各组用CPP箱进行成瘾性行为的检测。用PowerLab多通道生理信号采集与处理系统检测空白对照组、甲苯吸入组和醋酸吸入组大鼠的嗅球电位的改变。结果与其他各实验组相比,低浓度甲苯染毒组(328.6mg/m)和高浓度甲苯染毒组(1026.8mg/m3)有明显的CPP(P<0.01)。与低浓度甲苯染毒组比较,高浓度甲苯染毒组CPP更明显(P<0.05)。嗅球毁损的各实验组无明显的CPP。电生理学研究发现,空白对照组、甲苯吸入组、醋酸吸入组具有不同特征的嗅球电位。结论低剂量甲苯亚慢性吸入能够诱导大鼠的CPP行为,可能是通过嗅觉通路实现。
【Abstract】 Objective To know the effect of low level exposure of toluene on conditioned place preference (CPP) in rats. Methods 60 SD rats were randomly divided into 6 groups, 10 in each. 4 groups were treated respectively with toluene by inspiration of 328.6, 1 026.8mg/m3, OB-impaired+toluene 328.6 mg/m3, OB-impaired+1 026.8 mg/m3 in a closed chambers for 4 weeks (5 days/week, 6 hours/day) and 2 groups were taken as the control, OB-impaired control, blank control. The addiction and the generating electricity of the OB were examined by CCP chamber and the system of PowerLab. Results Compared with the blank group, in the toluene 328.6 and 1 026.8 mg/m3 groups the time in the light chamber was longer (P<0.01) and it is more longer in the 1 026.8mg/m3 group than that in the 328.6 mg/m3 group (P<0.05). OB-impaired group, OB-impaired+toluene 328.6 mg/m3 and 1 026.8 mg/m3 group showed no CPP. Electrophysiological examination showed that there was different OB potential in the groups. Conclusion Low level toluene exposure by inspiration can cause the CPP in rats, maybe the olfactory pathway is concerned.
