过氧化物酶体增殖物激活受体γ2基因多态性及其表达与糖尿病关系的研究
Relationship between peroxisome proliferator-activated receptor gamma2 Pro12Ala gene polymorphism and the expression of it in subcutaneous fat tissue and diabetes mellitus
【摘要】 通过PCR—RFLP与免疫组织化学方法,研究过氧化物酶体增殖物激活受体(PPAR)γ2基因Prol2Ala多态性与糖尿病(DM)易感性的关系,以及人皮下脂肪组织中PPARγ2的表达与DM的相关性.实验结果表明,DM患者基因型Pro/Ala为9%,而对照组Pro/Ala为5%,DM患者与对照组之间Prol2Ala多态性频率存在差异,但不显著;用SABC方法处理皮下脂肪组织后,DM患者与正常对照组切片染色不同.PPARγ2可能会在DM的诊断和治疗过程中起到一定作用.
【Abstract】 In this experiment,study the relationship between peroxisome proliferator-activated receptor(PPAR)γ2 gene Pro12Ala and the susceptibility of the diabetes mellitus(DM)as well as the connection between the expre-ssion of PPARγ2 in subcutaneous fat tissue and the DMby the PCR—PFLP and SABC method.The results demonstrated the genotype frequencies for Pro/Ala were 9% respectively in the DM group,while 5% in the control subjects.The Pro/Ala frequencies were not significantly higher in the patients than in the control group (P>0.05);After treating with the SABC method,there were differences in dye of tissue section between patients and control subject.The difference in dye displayed that the content of expression of PPARγ2 in subcutaneous fat tissue probably had the function of indication in diagnosis and treatment of DM.
