纤维蛋白原平板法测定水蛭素活性
THE FIBRINOGEN PLATE METHOD FOR ESTIMATING HIRUDIN ACTIVITY
【摘要】 目的:报告一种新的水蛭素活性测定方法——纤维蛋白原平板法。方法:以纤维蛋白原作为底物,在纤维蛋白原平板上进行免疫扩散反应,精确测量沉淀圈直径,绘制标准凝血酶活力标准曲线。在标准曲线上查出凝血酶与水蛭素反应后的剩余凝血酶活力,计算出水蛭素活性。结果:系列活力浓度10、5.0、2.5、1.25、0.625、0.3125NIH/ml的标准凝血酶其免疫扩散沉淀圈直径与酶活力呈良好的线性关系(r=0.9959)。结论:采用系列活力浓度的标准凝血酶与被水蛭素抑制后的剩余凝血酶同在一块纤维蛋白原平板上进行免疫扩散反应的方法,实验成本低,操作简便,结果客观,准确度较好。
【Abstract】 Objective:A new method, fibrinogen plate assay, for determination of the hirudin activity was reported in the paper. Methods:The immune pervasion reaction has been carried out on the fibrinogen plate with the fibrinogen as the substrate. The hirudin activity was evaluated by using the residual of activity of thrombin. After reacting with the hirudin, consultation was done on the standard curve about the thrombin activity obtained from the concentration dependence of the diameter of the deposit circles. Result: The diameter of deposit circles presents a linear relation with the thrombin activity (r=0.995 9) in the range from 10 to 0.312 5 NIH/ml. Conclusion: The method was found to be of advantages, such as low cost, easy operation and accuracy, for evaluating the activity of hirudin by carrying out the immune pervasion reaction of the standard thrombin and the residual of the thrombin reacting with hirudin on a same fibrinogen plate.
