骨髓瘤细胞系U266和急性髓性白血病细胞系KG1a细胞内砷浓度与细胞凋亡的关系

【摘要】 目的:探讨骨髓瘤细胞系U266与急性髓性白血病细胞系KG1a细胞内砷浓度与凋亡的关系。方法:用不同浓度三氧化二砷(As2O3)处理U266和KG1a细胞48h,原子吸收光谱法测定细胞内砷浓度,AnnexinV-FITC法检测细胞凋亡率。结果:随培养液As2O3浓度增加,U266细胞内砷浓度和凋亡率也增加,KG1a细胞无此现象。1.0μmol/LAs2O3处理时U266细胞内砷浓度高于KG1a细胞(P<0.01),2.0μmol/L As2O3处理时U266细胞凋亡率明显高于KG1a细胞(P<0.05)。细胞内砷浓度与凋亡率之间在U266细胞呈正相关(r=0.883,P=0.002),在KG1a细胞无相关(r=0.594,P=0.092)。结论:As2O3可诱导U266细胞凋亡而不能诱导KG1a细胞凋亡,对As2O3的敏感性U266细胞高于KG1a细胞,敏感性不同可能与细胞内砷浓度有关。

【Abstract】 Objective:To explore the relationship between intracellular concentrations of arsenic and apoptosis sensitivity in multiple myeloma U266 and acute myeloid leukemia KG1a cell lines.Methods:U266 and KG1a cell lines were cultured and treated with different concentrations of arsenic trioxide(As2O3)for 48 hours.Intracellular arsenic concentrations were assayed with atom absorption spectrum method.Apoptosis rate was evaluated with Annexin V-FITC flow cytometry.Result:The intracellular arsenic concentrations and apoptosis rates in U266 cells were increased as medium As2O3 concentrations were elevated.Such results were not seen in KG1a cells.When treated with 1.0 μmol /L As2O3,the intracellular arsenic concentration of U266 cells was markedly higher than that of KG1a cells(P<0.01).Treated with 2.0 μmol /L As2O3,U266 cells had significantly higher apoptosis rate than KG1a(P<0.05).Positive correlation between intracellular arsenic concentrations and apoptosis rates could be seen in U266 cells(r=0.883,P=0.002),but not in KG1a cells(r=0.594,P=0.092).Conclusion:As2O3 could induce apoptosis in U266 cell lines,but not in KG1a.U266 cell lines were more sensitive to As2O3 than KG1a.The difference in apoptosis sensitivity was probably associated with different intracellular arsenic concentrations.
