当归中阿魏酸的提取和纯化研究
Study on the Extraction and Purification of Ferulic Acid in Angelica Sinensis
【摘要】 以当归中主要活性物质阿魏酸的含量为指标,采用单因素及均匀设计实验的方法,确定了微波辅助提取当归中阿魏酸的最优工艺条件:乙醇浓度90%,微波功率850W,液固比6:1(mL:g),辐射时间9min,粒径大小为2mm,浸泡时间30min。结果表明:干浸膏中阿魏酸含量为0.212%,转移率为70.19%,均优于传统加热提取和超声提取方法,而且所用的时间分别是传统加热提取方法和超声提取方法的3/40和3/10。将高速逆流色谱技术应用于上述干浸膏中阿魏酸的分离纯化,经过一次分离得到的阿魏酸纯度为98.2%(HPLC)。HSCCC的溶剂系统组成为:正己烷-乙酸乙酯-甲醇-水(3:7:5:5)。用核磁共振和质谱表征了所得阿魏酸的化学结构。
【Abstract】 The ferulic acid is one of the main active components in Angelica sinensis(Danggui),and the microwave-assisted extraction was used for the extraction of ferulic acid from Danggui.The operation technological conditions of the microwave-assisted extraction were optimized by single factor experiments and uniform design method.The optimized technological parameters found are as follows:the 90% ethanol is used as solvent,the ratio of solvent to solid material is 6:1(mL:g),soak time is 30 min,the microwave irradiation time is 9 min and the microwave power is 850 W.Using the above optimum parameters,the extraction yield reaches 70.19% and the ferulic acid content in the dry powder obtained from the extraction of Danggui is 0.212%,these values are higher than that obtained by traditional heat-water(80℃)extraction and ultrasonic extraction method.The extraction time needed for microwave-assisted extraction is only 3/40 of that needed for heat-water extraction and is 3/10 of that needed for ultrasonic extraction method.Furthermore,using the high-speed counter-current chromatography(HSCCC)with the solvent system of n-hexane-ethyl acetate-methanol-water(3:7:5:5,v/v),the dry powder obtained from the extraction of Danggui was purified and the ferulic acid with purity of 98.2% was obtained.The purity mentioned was assayed by HPLC and its chemical structure was confirmed by using Mass and 1H-NMR.
