硫酸镁对大鼠MCAO模型血脑屏障及MAP-2表达的影响
Effects of magnesium sulfate on blood brain barrier and MAP-2 in Rats with MCAO
【摘要】 目的:观察硫酸镁对大鼠MCAO模型血脑屏障及神经细胞骨架结构微管相关蛋白-2(MAP-2)表达的影响,并探讨其可能的作用机制。方法:制备大鼠MCAO模型,并随机分为假手术组、单纯缺血再灌注组和硫酸镁治疗组,在梗死30min后硫酸镁治疗组静脉注射硫酸镁,其它2组静注生理盐水,梗死2h后再灌注,梗死后24h检测脑组织含水量、BBB通透性和MAP-2的表达。结果:与假手术组相比,单纯缺血再灌注组的脑组织含水量明显增加(P<0.01),BBB通透性显著升高(P<0.01),MAP-2表达明显下降(P<0.01)。与单纯缺血再灌注组相比,硫酸镁治疗组的脑组织含水量明显降低(P<0.05),BBB通透性显著性降低(P<0.05),MAP-2表达明显降低(P<0.05)。结论:硫酸镁能显著改善大鼠MCAO模型BBB的功能,减少MAP-2的降解,从而改善神经功能。
【Abstract】 Objective:To investigate effects of magnesium sulfate on blood brain barrier(BBB)and on expression of microtubule associated protein 2(MAP-2)in rats with MCAO.Methods:Cerebral ischemia-reperfusion model was generated by occluding the right middle cerebral artery(MCA)for 2 hours.The rats were randomly divided into sham operation,MCAO alone and treating groups.The treating group was treated with magnesium sulfate in 30 minutes after infarction,and the other 2 groups with normal saline(NS).In 24h after infarction,the content of water in brain,permeability of BBB and expression of MAP-2 were determined.Results:Compared with the MACO group without treating,the content of water in brain and the permeability of BBB in the treating group decreased remarkably(P<0.05).The expression of MAP-2 was obviously stronger than that in the MCAO group.Conclusion:Intravenous magnesium sulfate can improve neurologic function by increased the function of BBB and decreased the degradation of MAP2.
