水厂处理工艺中的微生物群落结构特征研究
Characteristics of Microbial Community Structures in Water Treatment Process
【摘要】 应用基因扩增—梯度变性凝胶电泳(PCR—DGGE)分子生物学方法对深圳市XL水库水源和BJ水厂处理工艺中的微生物群落结构特征进行了比较研究。结果表明,采用分子生物学方法能够比较全面地检测微生物群落,有利于准确评价饮用水的微生物安全性。16S rDNA鉴定表明,受污染的MK河水中含致病菌,存在微生物安全性风险,而在XL水库水和BJ水厂出水中均未发现致病菌,微生物安全性好。BJ水厂的处理工艺能够引起微生物群落结构的大幅度变化,水厂原水、砂滤池出水和活性炭池出水中的优势菌群几乎完全不同。掌握微生物群落特征的变化规律对于优化工艺运行具有指导意义。
【Abstract】 The microbial community structures in polluted MK river,XL reservoir and BJ water plant were studied with polymerase chain reaction/denaturing gradient gel electrophoresis(PCR/DGGE) molecular biology method.The results show that the microbial structural diversity in water samples can be fully revealed,the water safety in terms of microbiology can be evaluated.16S rDNA identification shows that there are pathogenic bacteria in MK river,which indicates lower microbial safety,while no pathoge-nic bacteria are detected in XL reservoir and BJ water plant,which indicates higher microbial safety.Water treatment process induces a remarkable change of microbial community structure in water.The predominant microorganisms are totally different among inlet water,sand filtration effluent and carbon filtration effluent.The results are of significance in guiding the optimal operation of water plant.
