生物催化肾上腺酮不对称合成R-肾上腺素
Asymmetric biosynthesis of R-adrenalin from corticosterone
【摘要】 研究了生物不对称还原肾上腺酮生成单一对映体R-肾上腺素.以肾上腺酮为前体,从土壤中筛选到一株对肾上腺酮具有较高立体选择性催化的菌株S289,以其为出发菌株,依次进行紫外和亚硝酸诱变,得到R-肾上腺素高转化菌株UN-78.确定了最佳转化条件,在初始底物浓度为10 mmol/L、摇床转速180 r/m in、温度30℃和pH 6.5,R-肾上腺素得率达56.2%,对映体过量值e达到99.8%.
【Abstract】 Asymmetric biosynthesis of R-adrenalin was studied in this paper.A new strain S289 was isolated from soil.This strain can synthesize R-adrenalin by asymmetric bioreduction of corticosterone,and it was selected as original strain.The mutant strain UN-78 with effective transforming ability and high enantioselectivity was obtained via ultraviolet and nitrous acid mutagenesis.After the optimization of transformation,the final yield and enantiomeric excess of R-adrenaline were 56.2% and 99.8% respectively,when the initial substrate concentration was 10 mmol/L,rotation speed 180 r/min,temperature 32 ℃ and pH 6.5.
