肝癌细胞黏附稳定性对其“重叠生长”的影响
Effects of Adherent Stability on the Overlapping Growth of Hepatic Tumor Cells
【摘要】 运用显微形态学观察、计算机图像处理技术和数据统计分析对细胞融合生长过程中的形变调节趋势进行定量表征;以细胞力学和生化检测等手段对细胞黏附力和整合素表达进行检测。结果发现:1)肝癌细胞运动变形较正常肝细胞强,其融合生长层内出现大量圆形细胞,且圆细胞多有聚集倾向;2)肝癌细胞株HepG2的Integrinβ1表达明显高于正常细胞株L02;3)"重叠"灶的圆细胞黏附力明显低于周边铺展细胞;4)纤维连接蛋白(fibronectin,Fn)对HepG2的Integrinβ1表达有下调作用;5)Fn裱衬后,HepG2黏附力增高,形变减弱,融合生长层圆细胞产生及聚集减少。由此推断:1)肝癌细胞整合素受-配体比对其细胞黏附稳定性有一定影响;2)肝癌细胞"重叠灶"的形成与细胞频发形变和大量圆细胞聚集存在相关性。
【Abstract】 Micro-morphological observation,Image Tool computer,analyzing systerm,statistical analysis,are use to process quantitative characterization on regulating cell morphological deformation in the growth of cell confluence.The experimental methods of cell mechanics and biochemistry is used to investigate the cell adhesion ability and the expression of integrin.As a result: 1)The ability of hepatoma cells to regulate cell morphological deformation was better than hepatic cells,so,there were many round cells produced in confluent hepatoma growth layer cells,and the round cells had a tendency toward getting together.2) The expression of Integrin β1 in HepG2 hepatoma cells was significantly higher than in LO2 hepatic cells.3) The adhesion ability of the round cells in the overlapping growth area was apparently weaker than that of confluent spreading cells.4)Fn down-regulated the expression of integrin β1 in HepG2 cells.5)When HepG2 cells were cultured on the Fn-coated surface,enhanced the cells’ adhesion ability,and reduced the morphological stability as well as the formation and aggregation of the round cells in confluent HepG2 growth layer cells.These results indicate: 1)The ratio of integrin receptor-ligand in hepatoma cells correlates with the cell adherent stability.2) The formed "overlaping growth plaque" in hepatoma cells relates to the cells’ high frequency morphological deformation and considerable amount of round cells’ aggregation.
