血管活性肠肽对大鼠脑缺血损伤的神经保护作用
Neuroprotective effects of vasoactive intestinal peptide against rat cerebral ischemia
【摘要】 目的探讨血管活性肠肽(VIP)对脑缺血损伤的神经保护作用和机制。方法SD大鼠侧脑室内注射VIP,通过管腔内线栓技术闭塞右侧大脑中动脉诱导建立暂时性局部脑缺血模型。采用TTC染色测定大鼠脑梗死体积;免疫组织化学染色观察S100β表达;双抗夹心ELISA检测血清S100β的含量。结果VIP注射后大鼠脑梗死体积较对照组明显缩小,减少约32.3%(P<0.05);S100β阳性细胞体积缩小,数目减少(P<0.05);血清S100β含量也明显降低(P<0.05)。结论VIP脑内注射可明显减小脑缺血后梗死体积,具有神经保护作用。VIP可以抑制S100β的过度表达,这可能是VIP的另一神经保护机制。
【Abstract】 Objective To explore the neuroprotective effect of vasoactive intestinal peptide (VIP) in rat ischemic brain injury. Methods VIP was administered via intracerebroventricular injection in SD rats prior to focal cerebral ischemia by intraluminal occlusion of the middle cerebral artery. The infarct volume was assessed with TTC staining, and immunohistochemistry was performed to analyze the S100β expression in the cerebral tissue, with the serum concentrations of S100β detected by double-antibody sandwich enzyme-linked immunosorbent assay. Results After VIP injection, the relative infarct volume in the rats with cerebral ischemia was significantly reduced by 32.3% as compared with the volume in the control group on day 1 (P<0.05), and the number of S100β-positive cells was significantly decreased in the cerebral tissue (P<0.05). The injection also resulted in significantly decreased serum S100β concentrations in the rats (P<0.05). Conclusion VIP injection can reduce the infarct volume in rats with focal cerebral ischemia, suggesting the neuroprotective effect of VIP in brain ischemia possibly by reducing S100β overexpression.
