食管鳞癌中zac基因的表达及临床意义
Expression of zac gene in esophageal squamous cell carcinoma and its clinical significance
【摘要】 目的:观察zac基因在食管癌中的表达及意义.方法:采用RT-PCR检测36例食管鳞状细胞癌组织,相应的癌旁组织和远癌组织中zac mRNA的表达情况.PCR产物经凝胶电泳,比较3种组织中zac与GAPDH条带的灰度值之比,半定量分析zac mRNA的表达水平.结果:36例食管癌组织中有14例(38.9%)zac mRNA检测到阳性表达,22例(61.1%)zac mRNA表达缺失,其阳性表达低于相应的癌旁组织(85.5%)和远癌组织(100.0%,P<0.05);食管癌组织中zac mRNA阳性表达水平(0.38±0.13)低于相应的癌旁组织(0.72±0.16)和远癌组织(0.80±0.12);随着肿瘤分化程度的升高,zac mRNA在食管癌组织中的阳性表达率也增加(P<0.05);zac mRNA阳性表达率在无淋巴结转移的癌组织中显著高于有淋巴结转移者(P<0.05),而临床病理分期和病理类型比较差异无统计学意义(P>0.05).结论:zac mRNA在食管癌组织中存在表达缺失和低表达,说明zac基因在食管癌的发生发展中可能具有抑癌基因作用,可能与食管癌的转移密切相关.
【Abstract】 AIM:To study the expression of zac gene in esophageal squamous cell carcinoma and its significance. METHODS: The expression of zac gene was detected by RT-PCR in 36 cases of esophageal cancer tissues, matched para-cancer tissues and tissues far away from tumor. PCR products were tested by gel electrophoresis. The expressions of zac gene in the three kinds of tissues were semi-quantified by calculating the ratio of zac and GAPDH gray value of bands respectively. RESULTS: The positive expression rate of zac mRNA in 36 cases of esophageal carcinoma was 38.8% (14/36), the negative expression rate of zac mRNA was 61.1% (22/36). The positive rate was lower than that in tumor-adjacent tissues (80.5%) and esophageal mucosa far away from tumor (100.0%), respectively (P<0.05). Semi-quantified analysis showed that the expression level of zac mRNA in esophageal cancer tissues (0.38±0.13) was lower than that in tumor-adjacent tissues (0.72±0.16) and esophageal mucosa far away from tumor (0.80±0.12). The higher positive expression rate of zac mRNA was correlated with higher differentiation grades in cancerous tissues (P<0.05). The positive rate of zac mRNA expression was higher in cancerous tissues without lymph node metastasis than in cancerous tissues with lymph node metastasis (P<0.05), but it was not correlated with TNM stages and histologic types of tumor (P>0.05). CONCLUSION: The expression of zac mRNA is lost or reduced in esophageal cancer tissues, which indicates that the zac gene may play a major role in suppressing the tumorigenesis and development in esophageal cancer, and that it may be correlated with metastasis of esopha-geal carcinoma.
