Leptin对Leptin受体在GH3细胞中表达的影响
Influence of leptin on expression of leptin receptors in GH3 cells
【摘要】 目的:观察Leptin受体(OB-R)在大鼠GH3细胞中的表达,研究Leptin对GH3细胞leptin受体表达的影响.方法:用RT-PCR方法检测Leptin受体3种亚型在大鼠GH3细胞中的表达,半定量PCR检测其表达的变化.结果:RT-PCR检测结果显示在大鼠GH3细胞上有Leptin受体3种亚型的表达,包括通用型受体OB-R,短型受体OB-Ra及长型受体OB-Rb,且leptin可上调大鼠GH3细胞Leptin受体mRNA的表达,其受体OB-Ra和OB-RbmRNA表达分别增加至(154±11)%和(188±9)%.结论:Leptin可能通过其在GH3细胞上的相应受体直接调控GH3细胞的功能,影响GH3细胞的生长、增殖和生长激素的分泌.
【Abstract】 AIM:To observe the influence of leptin on the expression level of leptin receptors (OB-R) in the cultured GH3 cells. METHODS: RT-PCR method was used to observe the expression of leptin receptors (OB-R) and its change in GH3 cells. RESULTS: There were three subtypes of leptin receptors mRNA expressed in GH3 cells, including OB-R (common form), OB-Ra (short form) and OB-Rb (long form). After leptin (10-8 mol/L) treatment for 48h, OB-Ra and OB-Rb mRNA levels in GH3 cells were increased to (154±11)% and (188±9)%. CONCLUSION: Leptin might regulate function of GH3 cells through leptin receptors in GH3 cells directly, including growth, proliferation and GH secretion of GH3 cells.
