bFGF-PLA-Ns对体外培养成骨细胞增殖、分化及矿化的影响
Effect of basic fibroblast growth factor-polylactide sustained release nanospheres on proliferation,differentiation and mineralization of osteoblasts in vitro
【摘要】 目的观察制备的碱性成纤维生长因子聚乳酸纳米微球缓释系统(bFGF-PLA-Ns)对体外培养的兔成骨细胞的增殖、分化和矿化能力的影响。方法体外培养兔成骨细胞并鉴定,将bFGF-PLA-Ns和第4代成骨细胞一起培养,碱性磷酸酶(ALP)试剂盒检测细胞ALP活性,茜素红染色方法观察矿化结节的形成,并与单纯bFGF和空白组的作用进行比较。结果bFGF-PLA-Ns具有良好的生物活性,能显著促进兔成骨细胞的分化和矿化,其效应高于单纯施加bFGF和空白组的效应。结论制备的bFGF-PLA-Ns比单纯的bFGF对体外培养的成骨细胞有更为显著的生物学效应,在骨创伤的治疗中有良好的前景。
【Abstract】 Objective To study the effects of prepared basic fibroblast growth factor-polylactide sustained release nanospheres(bFGF-PLA-Ns) on proliferation,differentiation and mineralization of the cultured rabbit osteoblasts.Methods New Zealand rabbit osteoblasts were cultured and identified.bFGF-PLA-Ns and bFGF were added into the medium of the fourth generation of rabbit osteoblasts.Alkaline phosphatase(ALP) activities were detected by ALP kit.The formation of mineral tubercula was showed by alizarin bordeaux staining.Results bFGF-PLA-Ns had biological activity.bFGF-PLA-Ns could significantly enhance the differentiation and mineralization of rabbit osteoblasts,with significantly higher effects than bFGF only and blank group.Conclusion The biological effects of prepared bFGF-PLA-Ns were significantly higher on cultured osteoblasts than that of common bFGF only and that of blank group,and bFGF-PLA-Ns have a better prospect in bone traumatic healing.
