茄红素-β-环化酶反义基因对胡萝卜茄红素含量的影响
Influence of Anti-Sense LYC-B Gene on Lycopene Synthesis in Daucus carota
【摘要】 以根癌农杆菌GV3101为介导,用茄红素-β-环化酶(lycopene bate cyclase-LYC-B)5′端反义基因对胡萝卜(Daucus carotaL.var.sativa)进行了遗传转化,经胚发生途径得到抗性植株。GUS检测和Southern杂交表明目的基因已整合到胡萝卜基因组中,Dot Blot显示目的基因在受体植株中已表达,茄红素和胡萝卜素检测结果表明,转基因愈伤组织茄红素含量高于胡萝卜素含量。
【Abstract】 Hypocotyls of carrot(Daucus carota L.var.sativa)were infected with Agrobacterium tumefaciens GV3101 containing anti-sense LYC-B fragment at 5′ end.The regenerated plants were obtained through somatic embryogenesis.GUS staining and Southern hybridization indicated that the target gene has been introduced into the genome of carrot.RNA dot blot showed that the introduced gene has expressed in transgenic plants.The lycopene content is higher than that of the carotene in the transgenic calli.
