人参果胶SB2-1的结构研究
Studies on structure of pectin SB2-1 from Panax ginseng
【摘要】 从我国名贵中药人参的根中分离纯化出人参果胶SB2-1,经高压液相色谱分析证明SB2-1为均一组分,相对分子质量约为7 000.气相色谱分析表明其单糖组成为GalA和Gal,物质的量比为3.43∶1.00.采用部分酸水解、果胶酶解、高碘酸氧化、Smith降解、甲基化分析和NMR分析等方法分析了SB2-1的结构,结果表明:SB2-1为少分枝结构,α(1→4)GalA构成主链的核心结构,(1→4)Gal位于主链的边缘,(1→4)GalA和(1→4)Gal在6-O处有分支,平均每20个己糖残基有1个分支,支链部分由(1→6)Gal和(1→3,6)Gal构成,末端残基为(1→)Gal,SB2-1中的主要糖苷键类型为α型.
【Abstract】 Pectin SB2-1 was isolated and purified from the root of Panax ginseng C.A.Mey.The HPLC analysis indicated that SB2-1 was homogenous.Its molecular eight was estimated by gel filtration to be 7 000.The GC analysis indicated thawt it contained the monosaccharides of GalA,and Gal.Their molar ratio is 3.43∶1.00.Partial hydrolysis with acid,pectinase treatment,periodate oxidation,Smith degradation,methylation analyses,GC/MS analyses and NMR analyses were used for the structure analyses of SB2-1.The results revealed that SB2-1 is of a lower branched structure.The core structure of the main chain is composed of α-(1→4)-linked-GalA,the border of the main chain is(1→4)-linked-Gal.Some of(1→4)-linked-GalA and(1→4)-linked-Gal residues are substituted at 6-O.On an average,there is one branch every twenty hexose residues.The side chain is composed of(1→6)-linked-Gal and(1→3,6)-linked-Gal.The nonreduced end is composed of Gal.The main glycosidic link of SB21 is α configuration.
