八氢番茄红素合成酶基因植物表达载体的构建及对人参的遗传转化
Construction of Plant Expression Vecto r with PSY Gene and Transformation of Ginseng
【摘要】 将八氢番茄红素合成酶基因重组于植物双元表达载体pBin438,得到重组质粒pBin438PSY.用冻融法将其导入农杆菌EHA101中,采用叶盘法转化人参愈伤组织,经诱导与筛选,获得了潮霉素抗性植株。提取抗性植株总DNA,通过PCR扩增和PCR-Southern杂交检测,筛选出了整合有外源基因的抗性细胞系。为进一步研究八氢番茄红素合成酶基因在人参中的表达及生物学功能的研究奠定了基础。
【Abstract】 The plant expression vector pBin438,carrying a PSY gene,was constructed.pBin438PSY was introduced in to Agrobacteriumtum efaciens by frozen-melting.Ginseng callus were transformed via Agrobactertum efaciens mediated pro-cedure.Transgenic Ginseng callus cells grew in induction medium and plants resistant to hygromycin were obtained.The PCR and PCR-Southern blotting analysis confirmed that PSY gene had integrated into the genome of tobacco.
