电离辐射对HepG2细胞系细胞周期的影响
EFFECTS OF RADIATION ON CELL CYCLE OF HUMAN HEPATOMA HEPG2 CELL LINE
【摘要】 目的:探讨放疗对人肝癌细胞系HepG2细胞细胞周期的影响。方法:以人肝癌细胞系HepG2细胞为研究对象:分别以2Gy、4Gy、6Gy、8Gy和10Gy吸收剂量的6MV-X线照射HepG2细胞,照射后培养24h;以4Gy吸收剂量的6MV-X线照射HepG2细胞,照射后分别培养6h、12h、24h和48h;以未予射线照射的同步培养细胞为对照。采用流式细胞技术检测不同照射剂量及照射后不同时间点HepG2细胞细胞周期的变化。结果:2Gy、4Gy、6Gy、8Gy和10Gy剂量6MV-X线照射后培养24h,HepG,细胞表现为随着照射剂量的增加,G1期细胞百分率下降;6Gy、8Gy和10Gy剂量组HepG2细胞表现为G2/M期阻滞。4GyX线照射后培养6h,即可观察到G2/M期阻滞,阻滞峰值为正常的56倍;然后细胞被释放,再度进入细胞周期或死亡。结论:不同剂量X线照射人肝癌细胞系HepG2细胞,大分割剂量组主要诱导G2/M期阻滞,4Gy剂量射线照射后,S期细胞阻滞明显,并与G2/M期阻滞同步解除,阻滞解除后启动增殖或进入凋亡。
【Abstract】 Objective:To investigate the effects of radiotherapy on cell cycle of human hepatoma HepG2 cell line.Methods:The human hepatoma HepG2 cell line was exposured to 6MV X-ray and the HepG2 cells without irradiation were taken as control.The HepG2 cells were respectively irradiated with 2Gy,4Gy,6Gy, 8Gy and 10Gy 6MV X-ray,after irradiation,the cells were all cultured 24h.The HepG2 cells were irradiated with 4Gy 6MV X-ray,after irradiation,the cells were respectively cultured 6h,12h,24h and 48h.Flow cytometry (FCM) was applied to detect the changes of HepG2 cells cell cycle of different dose of X-ray irradiation and at different culturing time after irradiation.Results:With increasing of X-ray dose,the percentage of G1 phase HepG2 cells decreased;The HepG2 cells were blocked at G2/M phase irradiated with 6Gy,8Gy and 10Gy X-ray. The HepG2 cells were observed blocking at G2/M phase when being cultured 6h after irradiated with 4Gy X- ray,and the blocking peak was 56 times higher than normal;Then,the cells entered the cycle again or died. Conclusions:The HepG2 cells were induced G2/M phase blocking after being irradiated with high fractionated dose X-ray;After the cells were being irradiated with 4Gy X-ray,the cells were obviously blocked at S phase and synchronously relief with G2/M phase,then start proliferation or apoptosis.
