盾叶薯蓣胚乳的组织培养

【摘要】 分别以盾叶薯蓣成熟种子和未成熟种子的胚乳为材料进行组织培养。结果表明:愈伤组织诱导培养中,无论是成熟胚乳还是未成熟培乳,采用MS+6-BA1.0mg.L-1+NAA0.1mg.L-1培养基诱导效果最佳,诱导率分别达到83.3%和90.0%;以1.0mg.L-16-BA+0.2mg.L-1NAA+MS培养基进行继代培养可达到比较理想效果;0.1mg.L-16-BA+0.05mg.L-1NAA+蔗糖3%+500mg.L-1CH+MS培养基诱导芽的分化,效果最好,成熟胚乳和未成熟胚乳芽诱导率分别为80.0%和90.0%。用1/2MS培养基进行生根培养时,ABT生根粉溶液处理的生根效果,在浓度200~300mg/L范围内没有显著差异,成熟胚乳再生植株生根率为76.6%,未成熟胚乳再生植株为83.3%。

【Abstract】 Tissue culture of endosperms from D.zingiberensis seeds was studied.The results were as follows:When mature endosperms and immature endosperms were dealt with MS+6-BA 1.0 mg·L-1+NAA 0.1 mg·L-1,both of them had a callus inducement percentage of 83.3% and 90% respectively.And the culture medium composed of 1.0 mg·L-16-BA+0.2 mg·L-1NAA+MS had excellent results in subculture.In the process of buds inducement,using 0.1 mg·L-16-BA+0.05 mg·L-1NAA+3%sugar+500 mg·L-1CH+ MS made the best result that there was a buds inducement percentage of 80% from calli of mature endosperms and 90% from calli of immature endosperms.In the process of generating root culture,the concentration of ABT rooting agentia had nothing with ratio of rooting in a range of 200~300 mg·L-1.The rooting ratio of endosperm calli of mature seeds was 76.6% and that of immature was 83.3% respectively.
