μ型阿片受体和催产素受体二聚化的研究
STUDY ON EXISTANCE OF DIMERIZATION OF μ OPIOID RECEPTOR AND OXYTOCIN RECEPTOR
【摘要】 目的探讨μ型阿片受体(MOR)和催产素受体(OTR)可否形成异源性受体二聚体。方法采用免疫共沉淀(IP)和荧光共振能量转移(FRET)方法,检测在MOR和OTR质粒共转染中国仓鼠卵巢(CHO)细胞和大鼠脑组织中是否存在MOR和OTR异源二聚体。结果IP方法检测显示,在MOR和OTR质粒共转染CHO细胞及大鼠脑组织中可见MOR和OTR蛋白条带。MOR和OTR共转染CHO细胞FRET方法检测信号为阳性(FRET率均值为2.01)。结论在MOR和OTR质粒共转染CHO细胞及大鼠脑组织中存在异源性MOR和OTR二聚体。
【Abstract】 Objective To investigate the existance of heterodimers of μ opioid receptor(MOR) and oxytocin receptor(OTR). Methods The immunoprecipitation(IP) and fluorescence resonance energy transfer(FRET) were used to study the dimerization of MOR and OTR in the Chinese hamster ovary cells(CHO) cotransfected by MOR and OTR plasmid and in the rat brain tissue. Results The bands of MOR and OTR were seen in the cotransfected CHO cells and rat brain tissue via IP test.The FRET signal was positive in the cotransfected CHO cells(average value of FRET rate was 2.01). Conclusion The heterodimers of MOR and OTR exist in the cotransfected CHO cells and brain tissues.
