高效液相色谱法研究氟尿苷二丁酸酯稳定性

【摘要】 目的:建立氟尿苷二丁酸酯的反相高效液相色谱分析方法,并对其稳定性进行研究。方法:用 Kromasil C18色谱柱(150mm×4.6 mm,5μm),甲醇-水(65:35)为流动相,流速1 mL·min-1,检测波长267 nm 等条件下分析测定氟尿苷二丁酸酯在不同 pH 磷酸缓冲盐溶液和小鼠组织匀浆中的降解情况,确定其稳定性。结果:方法平均回收率99.54%~102.50%,RSD<5.0%,回归方程为 A=1248.2+8751.4C,(r=0.9999)。结论:氟尿苷二丁酸酯在弱酸性溶液中稳定,在弱碱性溶液中水解较快;在组织匀浆中易水解,在血液中水解最快。

【Abstract】 Objective:A RP-HPLC method was established to determine floxuridinyl dibutylate and its stability was studied by this method.Method:The samples were determined on a Kromasil C18column (5μm,150mm× 4.6mm),with the mobile phase of methanol-water (65:35).The flow rate was 1mL·min-1,and the UV detec- tion was set at 267nm.The degradation of floxuridinyl dibutylate in different pH phosphate buffer solution and in homogenate of mice were determined.Results:The recoveries were 99.54%-102.50% ,RSD<0.1%.Regression equation was A=1248.2+8751.4C,(r=0.9999).Conclusion:Floxuridinyl dibutylate is stable in weak acid solu- tion and easily hydrolysed in alkalescence solution;It is easily hydrolysed in homogenate,most fastly hydrolysed in blood.
