大鼠血液样品中海鞘醇检测方法的建立
Method for assay of SC in rat plasma
【摘要】 目的:建立具有抗乙肝病毒活性的新化合物海鞘醇(5α,8α-环二氧-24-二甲基胆甾-6-烯-3β-醇,SC)的体内检测方法。方法:采用正相色谱,环己烷-丙酮(60:40)溶液为流动相,流速1.0 mL·min-1,蒸发光散射检测器,通过测定SD大鼠血浆中SC的含量来建立SC体内HPLC检测方法,并进行方法学的考察。结果:SC在3.2~64.0μg·mL-1范围内,呈良好二阶三项式关系,相关系数r=0.9999,回归方程为:Y=0.021X2-0.065X+0.287,血浆回收率试验证明,高低浓度回收率平均可达81.5%。结论:此方法稳定可靠,可以作为海鞘醇甚至其他甾醇类化合物的体内检测法。
【Abstract】 Objective:To establish the assay method in rat plasma of SC(5α,8α-cyclicobioxygen-24-bimethyl- 6-vinyl-3β-cholesterol)which possesses potential anti-HBV activities.Methods:Use positive phase chromato- gram,mobile phase cyclohexane-acetone(60:40),speed 1.0 mL·min-1,ELSD detector,by means of SC concentrations of SD rats plasma assay to set up SC HPLC measurement methods in plasma and put up method- ology examination.Results:When SC concentration range was 3.2-64.0μg·mL-1,recursive equation was Y=0.021X2-0.065X+0.287,r=0.9999,a bi-gradation trinomial.Average plasma recovery rate was 81.5 %.Conclusion:This method is stable and credible,it may be taken as an examination method in vivo to test SC or other steroids.
