毛细管区带电泳法测定人工种植粗毛淫羊藿中绿原酸的含量

【摘要】 目的:建立粗毛淫羊藿中绿原酸含量的 CZE 测定方法,比较不同地区人工种植粗毛淫羊藿中绿原酸含量。方法:采用熔融石英毛细管柱(50μm×95cm,有效长度86.8cm)作为分离通道;以20mmol·L-1硼砂(pH=9.12)为运行缓冲液;运行电压30kV;毛细管柱温20℃;检测波长:327nm;精密量取样品溶液,外标法测定含量。结果:粗毛淫羊藿中绿原酸成分得到完全分离,测定精密度得到明显改善;绿原酸的线性范围为3.28~42.64μg·mL-1,r=0.9995;方法的加样回收率为98.6%,RSD 为2.0%。结论:本方法简便、准确,重复性好。

【Abstract】 Objective:To establish a CZE method for determination of chlorogenic acid in Epimedium acuminatum Franch,comparing contents of chlorogenic acid in cultivated Epimedium acuminatum Franch in different sources. Method:A fused silica capillary(50 μm×95 cm,with effective length of 86.8 cm)was used.The running buffer was composed of 20 mmol·L-1 borax(pH=9.12).The applied voltage was 30 kV and the capillary temperature was 20 ℃.The detection wavelength was 327 nm.Sample solution was measured accurately,content was determined with external standard.Results:The peak of chlorogenic acid in Epimedium acuminatum Franch was separated com- pletely,precision was improved obviously.The linear curve for chlorogenic acid was obtained in the range of 3.28- 42.64 μg·mL-1,r=0.9995.The recovery was 98.6% with RSD of 2.0%.Conclusion:The method is simple,ac- curate and good in reproducibility.
